Chris McTurk scored a hat-trick of tries in the opening game, his side showed plenty of character after trailing 5-0 against Essex and John Gatus struck twice against Northumberland.
They now go forward to face Group one winners Middlesex in the quarter-finals, who reached the knock-out stages despite a 19-15 defeat at the hands of Somerset.
"We were kicking ourselves about that defeat by one score against Hertfordshire but today we’ve done a lot of good work and showed that we can score from anywhere," said coach Dave Rogers, from Wickersley School and Sports College.
"We made a lot of good decisions on the pitch and they’re starting to trust each other in defence and attack."
McTurk’s pace has seen him step up from Yorkshire’s B team this season and two young players who have caught the eye are Chris Walker (North Ribblesdale) and George Elliott (York), who has represented England at rugby league.
Other unbeaten sides going into the knock-out stages were Cheshire, Lancashire and East Midlands, who join group winners Surrey and Kent while Devon squeezed through as the best runner-up on tries scored.
Central to the weekend is the Development Festival, whose teams have been assisted by the SEEU@RUGB programme that helps take the game to new communities.
This year the inner-city teams have been joined by Lowestoft, who played three games on The Close, the birthplace of the game where William Webb Ellis picked up and ran with the ball.
Lowestoft won all four of their games on the opening day, beating Manchester, Nottingham, Birmingham and Leeds/Bradford, with tries from Alex Deank, Liam Colby and Scott Papworth.
"It’s an area of deprivation in terms of income, opportunities and education but these boys have come together and the way they have learned to support and work for each other is very good," said coach Bill Johnston, whose players from the Lynngrove and Cliff Park schools in Great Yarmouth.
"Only one of the squad plays rugby for a club – the rest are all footballers really – but they’re a pacy set of individuals who have performed really well so far."
Their main challenge on the second day looks likely to come from Essex and Liverpool, who both impressed with their combination of flair and raw talent.
